州 shows a sandbank within flowing water — the radical 川 (three-water-stroke form) plus dots for land. That “island in the river” broadened to mean a region or administrative area, hence uses like 本州 and 欧州.

Standard on-yomi is シュウ as in 本州 and 欧州. There is no commonly used kun-yomi in modern Japanese.
